Associate Scientist

Orna Therapeutics, Inc. seeks Associate Scientist for Watertown, MA.

Duties include: Plan and execute gram-scale productions of circular RNA or lipid nanoparticles for cross functional teams supporting development activities and in vivo preclinical studies; Plan and design experiments to assess scalable downstream process changes to optimize the clinical manufacturing process for increased product recovery and purity for both circular RNA and lipid nanoparticles (LNP); Mmanage relationships and projects with raw material and external research vendors by communicating relevant information and data to keep projects according to timelines; Compile and maintain database of internal and external manufacturing process data to trend collected data for circular RNA and LNP process understanding and present findings to cross functional team meetings.  Position requires BS or foreign equivalent in Biotechnology, Biochemistry, Chemistry, Bioengineering, Chemical Engineering/related field with 2 years of experience performing process development and internal manufacturing activities for RNA therapeutics, to include 1 year of lipid nanoparticle (LNP) therapeutic development. Experience must include the following, which may be gained concurrently:

  • 2 years’ experience performing bench milligram and gram scale circular RNA synthesis by in vitro transcription.
  • 2 years’ experience in performing downstream circular RNA purification methods including tangential flow filtration, affinity chromatography, and final filtration/fill at the gram scale.
  • 2 years’ experience executing and analyzing circular RNA using analytical assays such as ion pair reverse phase chromatography, spectrophotometry, pH, and osmolality.
  • 2 years’ experience manufacturing tox lot batches of circular RNA for IND enabling in vivo non-human primate studies.
  • 2 years’ experience analyzing data using GraphPad Prism and JMP software
  • 2 years of experience in process development to improve scalability, purity, and yield of circular RNA processes both upstream and downstream.
  • 2 years’ experience in process development documentation using electronic lab notebooks, writing protocols, and assisting with writing reports; along with maintenance of team’s raw materials and product inventory.
  • 1 year experience executing and analyzing LNP using analytical assays such as Ribogreen, and sizing by DLS (dynamic light scattering).
  • 1 year experience performing LNP-RNA formulations via Precision Nanosystem instruments including Ignite and Blaze.
